What do the numbers after the colon mean? e.g. tcpwm.line[0]:3.

This page confuses me when I start to configure pin functions.

How should I ensure sufficient pin resources at the beginning of design?

I want to assign each pin to specific functions but this datasheet doesn't seem to have a lot of useful information.

The description tcpwm.line:Y means

This pin can be connected to the line terminal of the Xth TCPWM block.

And this is the Yth pin where the TCPEM.line can be connected.

Is it hard for you to select a pin which can be connected to the line terminal?

Don't worry.  PSoC Creator provides a list of pins which can be used for the line terminal.
