I tried with the given example project "GPIOApp" to toggle the gpio21 pin, but i am not finding any changes.

With the original firmware and supposing your kit has no hardware problem, you will see GPIO 21 is toggling after booting.

I suggest you to check if the firmware reports any error message when intialize and boot.

Do you change anything in the example project? How do you observe the output of GPIO21? By an oscilloscope?

i changed gpio21 to gpio45 and in scope that pin in showing high
